The project works with the archaic image of the library as a "temple of knowledge" and at the same time, tries to shift its current institutional subtext, based on the idea of knowledge accumulation, towards its careful selection and interpretation. In earlier times, information was a scarce commodity and access to education was the privilege of a few social classes, closely connected to power. In the second half of the 19th century, the Enlightenment and the Industrial Revolution brought political and technological changes in the dissemination of information, that continue to accelerate. The machine printing of newspapers and books, the train, the telegraph, and later the telephone, as precursors to the digital dissemination of information, made it possible to transport information in quantities and at speeds previously unimaginable. Modern public institutions, public libraries among them, are emerging to make knowledge accessible to all. Today, however, it seems that it is not the desire for quantity of information and knowledge, but the desire and need to understand the world around us, that is immutable in human nature. The threshold, where being overwhelmed with information does not contribute to finding meaning and one's own position in the world, is now part of our everyday experience. The concept of a library based on the accumulation and easy retrieval of information seems redundant in the age of digital communication, and a principle that worked before may not work today. At the same time, the fact that libraries still work primarily with the medium of the physical book anchors the library in irrelevance. As a mediator of the image of the past, it ignores the present and its pressing issues. The medium of the book, on the other hand, can also be seen as the greatest privilege of libraries in the digital world, which however makes them a different institution. In this project, I therefore propose a new concept of the library as a centre of information management, whose aim is to make information available in a valid way. Librarians, as information specialists, are both curators of the institution's content and of the way it is "exposed" to the public. The library therefore adopts the principles of other Enlightenment institutions such as the museum and the gallery. Librarians/curators are working with a particular topical theme that culminates in its publicly available organized catalog during its exposure. The result is a curated selection of information on that topic - a collection presented in an appropriate manner that reflects current expectations and world views. An apparatus that responds to the present and underpins it with the past. A place to pause and reflect in-depth on a defined topic.
Wingless/Notch crosstalk in Drosophila melanogaster

Základním kamenem existence mnohobuněčných organismů je přítomností signalizačních drah, komplexních biochemických kaskád umožňujících vzájemnou komunikaci mezi buňkami. Dráhy Notch a Wnt jsou klíčové pro správný embryonální vývoj, a jejich deregulace vede k vzniku mnoha závažných onemocnění. Mnohaletý výzkum postavený z velké míry na poznatcích z pokusů v modelovém organismu D. melanogaster ukázal, že komponenty těchto drah nepůsobí striktně odděleně, ale naopak vstupují do vzájemné interakce (crosstalk). Tento fenomén je však u savců prozkoumán jen v omezené míře. Tato bakalářská práce má za cíl shrnout dosavadní poznatky o spolupůsobení drah Wnt a Notch u D. melanogaster a nastínit, kde by mohl crosstalk hrát roli u savců.
Pathways Identification In The Gram-Positive Bacterium Aneurinibacillus Species H1

Here, we present the first insight into the Gram-positive, promising polyhydroxyalkanoatesproducer Aneurinibacillus species H1. Both static and dynamic properties are described inthis paper with the aim on identification of pathways occurring in the organism. The genome consistsof a circular, 3,663,644 bp long chromosome and contains 4,654 protein-coding sequences and129 RNAs in total. The GC content is 44.8%. Functional properties identification showed that CDSdivide into 26 categories with the most prevalent Amino Acids and Derivatives group. Pathwaysinference revealed 201 pathways. The most represented group is metabolic pathways, which is furtherdivided into 12 groups.
Evolution of the Heme Biosynthetic Pathway in Eukaryotic Phototrophs

This thesis is devoted to the evolution of the heme biosynthetic pathway in eukaryotic phototrophs with particular emphasis on algae possessing secondary and tertiary red and green derived plastids. Based on molecular biology and bioinformatics approaches it explores the diversity and similarities in heme biosynthesis among different algae. The core study of this thesis describes the heme biosynthesis in Bigelowiella natans and Guillardia theta, algae containing a remnant endosymbiont nucleus within their plastids, in dinoflagellates containing tertiary endosymbionts derived from diatoms called dinotoms, and in Lepidodinium chlorophorum, a dinoflagellate containing a secondary green plastid. The thesis further focusses on new insights in the heme biosynthetic pathway and general origin of the genes in chromerids the group of free-living algae closely related to apicomplexan parasites.

Metabolické toky biosyntetickými drahami aminokyselin s větveným řetězcem u Corynebacterium glutamicum mohou být modulovány různými změnami exprese genů příslušných drah i drah konkurenčních. Cílená mutagenese promotorů příslušných genů je vhodným prostředkem pro dosažení požadovaných změn genové exprese.
